MOTS-c

MOTS-c was discovered in 2015 by the lab of Changhan Lee at the University of Southern California. It is one of the few known peptides encoded not in the cell nucleus but in mitochondrial DNA, and it switches on AMPK, the cell's master metabolic switch. The effect resembles aerobic exercise, which is why MOTS-c is known as the exercise mimetic.In the original study in Cell Metabolism, MOTS-c protected mice on a high-fat diet from weight gain and insulin resistance. A 2021 study in Nature Communications showed that its level in human muscle rises 11.9-fold with exercise, and that treatment doubled running capacity in old mice.Natural MOTS-c levels decline with age, and a variant of the MOTS-c gene has been linked to exceptional longevity in studies of Japanese centenarians.
