Overview
Tirzepatide is a dual GIP and GLP-1 receptor agonist developed by Eli Lilly. It is the active molecule in Mounjaro (FDA approved in 2022) and Zepbound (FDA approved in 2023 for chronic weight management). By working on two incretin pathways at once, it acts on appetite, the feeling of fullness and how the body handles glucose after a meal.In the SURMOUNT-1 trial, 2,539 adults with obesity or overweight took part for 72 weeks. Participants on the highest dose lost 20.9% of their body weight on average, against 3.1% on placebo. In the head-to-head SURMOUNT-5 trial, tirzepatide reached 20.2% average weight loss versus 13.7% for semaglutide.With a half-life of about five days, it was studied as a once-weekly injection.
