Overview
Retatrutide (LY3437943) is an investigational once-weekly molecule developed by Eli Lilly. It adds a third target to the GIP and GLP-1 pair: the glucagon receptor, which is linked to energy expenditure and fat metabolism in the liver. Three pathways in one molecule is what sets it apart from every approved incretin drug.In the Phase 2 trial published in the New England Journal of Medicine (2023), 338 adults with obesity took part for 48 weeks. Participants on the 12 mg dose lost 24.2% of their body weight on average, against 2.1% on placebo. The Phase 3 TRIUMPH programme went further: according to Lilly, 28.7% at 68 weeks in TRIUMPH-4 and 28.3% at 80 weeks in TRIUMPH-1.Retatrutide is still in late-stage trials, which makes it one of the most closely watched molecules in metabolic research.
